01 / OVERVIEW

What is it?

A preparation of water-soluble polypeptide fractions extracted from bovine pineal glands, manufactured by Geropharm with glycine as a stabilizer. It is a tissue-derived mixture, not a single defined peptide or melatonin itself.

Identity / background source ↗

Research context

A bovine pineal-gland polypeptide mixture investigated in menopausal symptoms and age-related melatonin physiology. It is distinct from Pinealon (EDR), synthetic Epitalon (AEDG), and the separately studied extract Epithalamin.

Selected study findings

randomized trial · 2016

Study of this preparation · Original clinical report

120 postmenopausal women aged 45–60; placebo 30, one-course group 30, two-course group 60

The multicenter double-blind trial reported improvement in modified Kupperman symptom scores versus placebo, principally neurovegetative symptoms. The two-course group retained a reported difference at day 180. Nineteen participants withdrew; 101 comprised the per-protocol population.

Limit: The report recorded 22 adverse events; normal laboratory measures do not prove overall safety. Allocation concealment, missing-data handling and clinical importance of the score differences need detailed appraisal. Related 2015/2016 reports may describe the same cohort; do not count them as independent trials. Menopause symptom findings do not establish general insomnia treatment or longevity effects.

observational · 2017

Study of this preparation · Original human biomarker report

55 older patients with reduced pineal melatonin function

The Russian abstract reports a 1.9-fold increase in nighttime urinary 6-sulfatoxymelatonin compared with the pretreatment value. This is a melatonin-metabolite finding, not a measured improvement in sleep architecture.

Limit: Randomization and a concurrent placebo group are not established by the abstract. The comparison with Epithalamin refers to earlier findings, not an established head-to-head trial. No demonstrated REM-sleep, lifespan or healthy-person performance benefit follows from this biomarker. Author affiliations include Geropharm; independent confirmation requires assessment.

02 / EVIDENCE DIMENSIONS

Evidence assessment

Human research
—Placebo-controlled menopausal-symptom trial and an older-adult biomarker report
Study methods
—Clinical symptom scales and urinary melatonin-metabolite measurement address different questions
Independent replication
—Independent replication and overlap between menopause publications require assessment
Source access
—Publisher trial report, PubMed abstract and manufacturer identity information
Safety evidence
—Short clinical follow-up does not establish uncommon harms or long-term safety

These fields describe different aspects of the evidence. A missing record does not mean that no research exists.

Outstanding review questions

Check full allocation, comparator, prespecified outcomes, effect estimates, harms and shared-cohort reporting before drawing a clinical conclusion.

Human reports are linked; their presence does not establish effectiveness. Publication records can share participants. Source counts are not trial counts.
